Tth DNA polymerase is a thermostable enzyme of approximately 94kDa isolated from Eubacterium Thermus thermophilus strain HB8. This enzyme replicates DNA at 74°C. It reveals RNA-dependent DNA polymerase activity in the presence of Mn2+ ions. The Conc. of RNA template for effective reverse transcription with Tth DNA Polymerase should be higher than for reaction directed by reverse Transcriptases(M-MuLV, AMV).
SPECIFICATIOS : Enzyme is supplied at a Conc. of 5U/µl along with one vial of 10X PCR buffer, one vial of 5 X RT-PCR Buffer, one vial of RT buffer 10X,one vial of 25mM MgCl2, &, one vial of additional 25mM MgCl2 & one vial of DNA's Free water.
STORAGE BUFFER : 10 mM TrisHCL (pH 7.5), 300 mM KCL, 0.1mM EDTA, 1mM DTT, 50% glycerol.
SUPPLIED REACTION BUFFERS :
(10X) PCR Buffer : 100 mM Tris HCL(pH 8.9), 1M KCL, 500g/ml BSA, 0.5% Tween 20 and 15 mM MgCl2
(5X) RT/PCR Buffer : 250 mM bicine/KOH(pH 8.2) 570 mM K-acetate 40% Glycerol(v/v)
(10X) RT Buffer : 100 mM Tris HCL(pH 8.9) 900 mM KCL.
Recommended Conc of Mg2+ 3-6 mM & Mn2+ 1-2mM for RNA dependent cDNA synthesis.
STORAGE : Recommended storage is at -20°C.
APPLICATIONS : cDNA Synthesis, single tube RT/PCR, Amplification of DNA Fragments of varied sizes ranging from about 100 bp to 3 kb by PCR.
